The son of Robert Slaughter & Mary, the daughter of Augustine Smith; Col. Slaughter served the country as Ensign and Lieutenant in the French & Indian Wars. A member of the Committee of Safety for Culpeper, vestryman & church warden of St. Mark's Parish and justice of the peace; he led a regiment in the Battle of Great Bridge, the first engagement of the Revolution on Virginia soil.

In 1756 he had married Susannah, the daughter of Mjr. Philip & Ann (Coleman) Clayton. In June of 1767 James purchased 200 acres of the farm along Mountain Run in Culpeper of Sam & Ann (Coleman) Clayton, his brother & sister-in-law.

Their children were: first a son, Capt. Philip Slaughter who married Peggy Margaret French (Strother) by whom he had six children; Lucy who married Isaac Hite Williams, Daniel F.,Susan, Mary A., Sally, and Eliza F. who married Col. John Dade. Following his first wife's death, Capt. Philip Slaughter then married the widow Brock, Elizabeth (Towles), on the 19th of January 1803. Of this union came: Dr. Thomas Towles Slaughter, the Rev. Philip Slaughter; Ann Mercer (Slaughter) Slaughter & Mary Smith (Slaughter) Thompson.

A daughter, Mary "Polly" who married John Miller Bell; a son Robert who married Margaret Bowie Pendleton; a son Judge James who married Margaret Strother (Gray); son Col Samuel Slaughter who married Frances Bruce Banks, the mother of Col. Wm. Banks Slaughter who organized the State of Wisconsin; and after her death, Virginia Stanard; son George who married Teresa Kelso; daughter Anne who married Reuben Fry; son Thomas who married Lucy Booker Bibbs; daughter Susan who married Judge Thomas Speed of Bardstown, Kentucky; and daughter Sarah who married James H. McLaughlin.
