39°39′31″N 83°44′18″W (39.658604, -83.738453)[3]. Parts of Jamestown are located in Silvercreek, Ross, and New Jasper Townships.
According to the United States Census Bureau, the village has a total area of 1.2 square miles (3.1 km2), all of it land.
Jamestown is located south of US Route 35 on State Highway 72. It is east of Lake Shawnee.

The area was first surveyed around 1815.
