Samuel Breneisen, Sr. was the son of Valentine Breneisen II and his wife, Salome Herman. Trinity Lutheran Church, Lancaster, PA, records indicate that Samuel was baptized on May 24, 1772. He married Susanna Barbara Schwartz, daughter of George and Elizabeth (Nein) Schwartz on February 18, 1799.

Samuel and Susanna Breneisen lived in Reading (Berks County), and then lived both in Adamstown and Reamstown (Lancaster County). Samuel and Susanna were innkeepers at the Rogers Tavern in Adamstown. He was also a reputable clockmaker and in 2016, one of his lovely clocks was on display at the Theodore R. Sprecher Museum in Ephrata, PA.
