The following children have been attributed to John Comstock:
Samuel 1715-1755 - m. Anne Brown
Joseph - believed to have died young
Jeremiah - d. 1791 - m. Phebe Arnold
John Jr. - born abt. 1716 / d. 1813 - m. Mary Ballou
Jonathan - 172-=1800 - married twice - 1) Sarah Comstock (a cousin) 2) Susannah Comstock Matteson (sister of his first wife, Sarah)
James - 1733- 1791 - m. Esther Comstock
Nathan - 1735-1816 - m. Mary Staples
Ichabod 1734-1800 - m. Sarah Jenckes (daughter of John Jenckes)
Esther - 1727-1785 - m. Peter Aldrich - Peter Aldrich was a Revolutionary War soldier
Based on the year of death for Esther Jenckes and the birth dates of Samuel, John and Esther - it would appear these children were the product of her marriage to John Comstock.
It is further likely that children born to John Comstock after Esther's death are likely the children of John and his second wife Sarah Dexter.
Unfortunately, not all references to the children of John Comstock are consistent in birth/death dates - nor which children were born to each of his wives.
